A synopsis clearly describes your manuscript's plot, character, setting, style, and mood. It demonstrates your writing style and shows your ability to craft a good story, and above all else, it should get editors, agents, and publishing house clamoring to read your full manuscript. 

A query letter is a one-page letter that masterfully sells your manuscript. It is your calling card. If you write it poorly, an agent/publisher will assume your book is also poorly written. It must stimulate and intrigue to secure a further look into your work.
